Core Promoter

• Region 50-150 base pairs upstream from the transcription start site which includes the TATA box.
• This core promoter is unable to mediate transcription by itself.  It needs promoter-proximal elements.

Promoter-Proximal elements

• CCAAT box is a cis-acting sequence
• similarly with the GC rich segment
• There are consequences for transcription when these sequences are mutated
